实验方案
- 样本准备：分离外周血单个核细胞，并重悬于分离缓冲液中。
- 标记：向样本中加入分离cocktail，在室温下孵育5分钟。
- 磁珠孵育：加入抗生物素纳米磁珠，在2-8°C下孵育10分钟。
- 分离：将细胞悬液加入置于磁场中的LS柱。收集流出液，其中含有富集的、未受影响的CD4+ T细胞。

Efficient Enrichment of High-Purity CD4+ T Cells from Human PBMCs Using SOLIDEX®-ISOEx Untouched 人CD4+ T细胞 Isolation Kit （柱式）

To evaluate the isolation efficiency of the SOLIDEX®-ISOEx Untouched 人CD4+ T细胞 Isolation Kit （柱式）, CD4+ T cells were enriched from human peripheral blood mononuclear cells (hPBMCs) via negative selection. Non-target cells were labeled with a biotinylated antibody cocktail and depleted using magnetic beads within a separation column.
Cell fractions were stained with CD3-APC and CD4-PE for flow cytometric analysis. The frequency of CD4+ T cells demonstrated a substantial increase from 23.3% in the pre-isolation sample to 94.4% in the post-isolation fraction. These results confirm that the 柱式 system provides superior depletion of non-target populations, yielding highly purified "untouched" CD4+ T cells.
